 The aryl hydrocarbon receptor (AhR) is a ligand-activated transcriptional factor. 2,3,7,8-tetrachlorodibenzo-p -dioxin (TCDD) is high affinity and toxic to many vertebrate animals. For instance, in many species, administration of TCDD and related compounds produces hepatomegaly, thymus atrophy and wasting syndrome. In mammalian, TCDD treatment produces hyperplasia, hyperkeratosis and works on promotion process of tumorigenesis. The toxicity of pollutants has been evaluated mainly according to their ability to activate the AhR system. Therefore, the binding affinity of pollutants and the induction of target genes are related to their toxicity. It has been proposed that inactive AhR remains in the cytoplasm and makes a complex containing two molecules of Hsp90, XAP2 and p23 in the absence of ligand. Ligand binding to AhR induces conformational change in the AhR undergoing translocation into the nucleus. In the nucleus, AhR associates with ARNT, partner protein in nucleus, recognizes xenobiotic responsive elements (XRE) containing core sequence GCGTG within the regulatory region, and induces transcription of the target genes, including Cytochrome P450, 1A1 (Cyp1a1 ), -1A2 (Cyp1a2 ), -1B1 (Cyp1b1 ), NADPH: quinine oxidoreductase 1 (Nqo1 ) and others. Although AhR homologs have been found in invertebrate species, for example Drosophila melanogaster AhR (Spineless) and Caenorhabditis elegans AhR (AhR-1), characteristics of invertebrate AhRs are different from the vertebrate proteins. In fact, AhR protein of Drosophila, C. elegans and soft-shell clam cannot bind exogenous ligands. However, Drosophila AhR does form a heterodimer with the Drosophila ARNT homolog (Tango) and activates transcription without binding a ligand.

 What controls the inherent differences of functions of AhR between vertebrate and invertebrate? I questioned whether key amino acids exist in PAS domain or some factors affect on ligand dependency. In this paper, I used chimeras between mouse and Drosophila AhR (mAhR and dAhR) to analyze the amino acids involved in ligand-dependency. I constructed six chimeras in total by exchanging three domains (DNA-binding, ligand-binding, and transactivation domains). The AhRs that contained the Drosophila ligand binding domain (LBD), such as MDM, DDM and dAhR, were constitutively active. However, the transcriptional activity of DMM, which contained the mouse LBD and transactivation domain (TAD), was ligand-dependent in S2 cells, and the results were similar to those obtained in Hepa-1c1c7 cells. The chimeric AhR revealed that the LBD determines constitutive transactivation in dAhR or ligand-dependent activation in mAhR. The LBD was further divided into three blocks that corresponded to amino acids 230-300, 301-361, and 361-420 of the mouse sequence. Analysis of six chimeric proteins clarified that amino acids 291-350 of the Drosophila LBD were required to keep the protein in the active form in the absence of ligand binding, whereas in the mouse AhR, this region was required to maintain the protein in the inactive form in the absence of ligand.

 Given that the LBD is important for ligand-dependent activity, I tried to identify which amino acids in the LBD, especially in the 300-361aa region, play an essential role. Previous reports have identified the key amino acid residues in the LBD of mAhR as Ile319, Cys327 and Ala375. These amino acids are conserved reasonably well among vertebrate animals. The corresponding amino acids in dAhR were assigned as Val308, Val316 and Cys364, respectively, and they were located in a hydrophobic pocket. I searched for amino acids that were conserved among vertebrate AhRs, but were not conserved in the Drosophila and C. elegans proteins. I selected four amino acid residues, Gly293, Arg312, Ala361 and Gln377, in the mAhR, which were in the region of the hydrophobic pocket. The S283G, +Arg, C316C, C364A and H366Q mutations decreased the level of transactivation of the reporter gene compared with that of wild-type mAhR. However, these mutations did not change the ligand-dependency from dAhR- to mAhR-type. The V308I and S350A mutation did not affect transactivation activity. Furthermore, site-directed mutagenesis revealed that Arg346 in the mouse LBD is an important amino acid that was critical for AhR activation.

 The inactive AhR makes a complex containing two molecules of Hsp90, XAP2 and p23 in the absence of ligand. How chimeric AhR series make a complex? I investigated complex formation of each chimeric AhR by using native PAGE. The band in low molecular weight (L) of MDM and mdm comlex probably represents preferentially-activated form which is insufficient complex and tends to change into active form. However, both high (H) and low (L) molecular weight bands of dAhR were not detected in the presence of ligand. In the ligand-dependent activation, AhR may be fully dissociated from the complex and localized into nucleus by ligand binding. Previously, several studies have reported that AhR associates with Hsp90 by the method of anion-exchange chromatography and sedimentation analysis of cytosol incubated with [3H]TCDD. In the presence of agonist, both 9S and 6S complexes was detected. The 9S cytosolic form of the AhR has been shown to be associated with Hsp90 whereas the 6S form has been shown to be Hsp90-free. H band and L band may correspond to 9S and 6S, respectively. Hsp90 plays a role in inactive form of AhR complex. The chimeric AhR/dmd was deficient in dissociation from the complex and was constitutively inactive. This result raises the possibility that N (230-299 aa) and C (362-420 aa) regions are important in the dissociation of AhR/dmd from the complex. These regions probably may play a key role in conformational change to dissociate from the complex. Further analysis will be required to determine whether these chimeric AhR can bind the ligand.

 In conclusion, in vertebrate, the role of ligand binding for AhR is the dissociation from the complex and the transformation into active form. In invertebrate, AhR constitutively maintains active form and this property may be attributable to the insufficient association with Hsp90 and co-chaperone. These results may explain the species difference of the binding ability of AhR with dioxin compounds. Furthermore, the results support the role of AhR as a sensor of extracellular signals that induces biological responses, and the conformational change of AhR in the active or inactive form.

 芳香族炭化水素受容体(AhR)は,リガンド依存的に活性化される転写因子である. 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)は,親和性が高く,多くの脊椎動物において毒性がある. 例えば,多くの種において,TCDDとTCDDに関連した化合物の投与は肝腫,胸腺の委縮と消耗性症候群を引き起こす. 哺乳類においては,TCDD処理は過形成や角質増殖を引き起こし,発癌のプロモーション過程に働く. 汚染物質の毒性は,主にAhRの活性化システムに依存していることが明らかになってきた. そのため,汚染物質の結合親和性と標的遺伝子の誘導は,毒性と関連がある. これまで,リガンドの非存在下において,不活性なAhRは細胞質に留まり,2分子のHsp90とXAP2,p23で構成された 複合体を形成していることが分かっている. AhRのホモログは,無脊椎動物にも見つかっており,例えば,Drosophila melanogaster AhR (Spineless)や Caenorhabditis elegans AhR (AhR-1)が挙げられるが,特徴は脊椎動物と異なる点がある. 実際,Drosophila, C. elegans と soft-shell clamのAhRは,外来性のリガンドと結合しない. しかしながら,Drosophila AhRは,Drosophila ARNT ホモログ(Tango)と結合し, リガンド結合することなく転写を活性化する.

 脊椎動物と無脊椎動物の間で,AhRの機能性の違いは何によって調節されているのだろうか? PASドメイン内に鍵となるアミノ酸が存在するのだろうか,それとも何か別の因子がリガンド要求性に影響を与えているのだろうか? この研究で,リガンド要求性に関与するアミノ酸を解析するために,マウスとショウジョウバエのAhR(mAhRとdAhR)のキメラを用いた. 3つのドメイン(DNA結合ドメイン,リガンド結合ドメイン,転写活性ドメイン)を入れ替え,全部で6つのキメラを作製した. MDM,DDM,dAhRのようなショウジョウバエのリガンド結合ドメイン(LBD)を持つAhRは,恒常的に活性化した. しかしながら,mAhR,DMMのようなマウスLBDとマウス転写活性ドメイン(TAD)を持つAhRは, S2細胞,Hepa-1c1c7細胞においてリガンド依存的な活性を示した. キメラAhRを用いた解析により,LBDはショウジョウバエにとって恒常的な活性化に, マウスにとってはリガンド依存的な活性化に重要であることが明らかになった. LBDをさらにマウスの配列で230-300, 301-361, 及び 361-420に相当する3つのブロックに分割し,6つのキメラタンパクを作製した. ショウジョウバエLBDの291-350番目のアミノ酸配列がリガンド非存在下における活性型を維持するために必要であること, 一方で, マウスAhRにおいてこの領域はリガンド非存在下における不活性型の維持に必要であることを明らかにした.

 LBDがリガンド依存的な活性に重要であることから,LBD内の特に300-361番目のアミノ酸配列で重要な役割を持つアミノ酸を同定しようと試みた. 以前の報告により,Ile319,Cys327,Ala375がmAhRのLBDにおいて重要なアミノ酸であることが同定された. これらのアミノ酸は,脊椎動物間でよく保存されたアミノ酸である. 一方で,dAhRにおいて,それぞれVal308,Val316, Cys364であり,さらに,疎水性ポケットに位置している. 他にも脊椎動物で保存され,DrosophilaC.elegans で保存されていないアミノ酸を探した. mAhRにおいて疎水性ポケットに位置する4つのアミノ酸Gly293, Arg312, Ala361と Gln377を選択した. S283G, +Arg, C316C, C364AとH366Qの変異体は,mAhRと同様の活性を示した. しかしながら,これらの変異体はdAhR型からmAhR型へのリガンド要求性の変化を引き起こさなかった. V308IとS350A変異体は,転写活性に影響しなかった. また,さらなる部位特異的変異導入法は, LBD内のArg346がAhRの活性化に重要なアミノ酸であることを明らかにした.

 リガンド非存在下において,不活性なAhRはHsp90,XAP2,p23と複合体を形成することから, キメラAhRはどのような複合体を形成しているのか, 作製したキメラAhRについてそれぞれネイティブページにより複合体形成について解析した. MDMとmdmの低分子の位置に検出されたバンド(L)は,おそらく優先的に活性化される状態であり, 複合体形成が不十分なために活性型に変化しやすい状態にあると考えられる. しかしながら,dAhRはH(高分子の位置で検出されたバンド)とLのバンドがどちらも検出されなかった. リガンド依存的な活性化において,リガンド結合によってAhRは複合体から解離し,核へ移行する. 以前,細胞質画分と[3H]TCDD を反応させ,anion-exchange chromatographyとsedimentation analysisを用いた研究が報告された. リガンド存在下において9Sと6Sのピークが検出された. AhRの9Sは,Hsp90結合型を示し,6SはHsp90解離型を示した. このことから,HバンドとLバンドは,それぞれ9Sと6Sに相当すると考えられる. Hsp90はAhR複合体の不活性型に維持する役割を持つ.キメラAhR/dmdは複合体から解離することができず,恒常的に活性がない. このことから,N(230-299)とC(362-420)は複合体から解離するための重要な機能を担っている可能性が示唆された. これらの領域はおそらく,複合体から解離するための構造変化に重要な働きを持っていると考えられる. 今後,キメラAhRが直接リガンド結合するかどうか確認するためのさらなる解析が必要である.

 以上のことをまとめると,無脊椎動物において,AhRは恒常的に活性型が維持されているが, これはおそらくHsp90やシャペロン補助因子との不十分な結合に起因することが考えられる. これらの結果は,種間のダイオキシン化合物の結合に対する相違点を説明していると考えられる. さらに,生物学的な応答を誘導する細胞外シグナルのセンサーとしてのAhR役割を裏付け, AhRの活性型や不活性型における立体構造の変化を支持する結果を得ることができた.
