Thermogenesis is a phenomenon in which the temperature of a specific floral tissue increases due to endogenous heat production. Over 200 years ago, thermogenesis in plants was first described by the French biologist, Jean-Baptiste de Lamarck. To date, thermogenesis has been reported in several species of the arum lily family, and also in palms, lotus, cycads, and water lilies. Among the thermogenic plants that have been thus far characterized, the sacred lotus and skunk cabbage are known as homeothermic plants, and are capable of increasing and maintaining their inflorescence temperatures for several days. For instance, the temperature of the lotus receptacle can be maintained at between 30-36℃ during a 2-4 day period, despite changes in environmental temperatures of between 10-45℃. In addition, the spadix of the skunk cabbage, which belongs to the aroid family, can generate and maintain an internal temperature of about 20℃ even when the ambient air temperature drops below 0℃ Temperature and respiratory measurement of thermogenic sites, such as inflorescence and receptacle,
has been carried out extensively, however, little is known about the molecular mechanism of thermogenesis in plants.
Also little is known about the involvement of plant UCPs in thermogenesis.
In this thesis, I focused on both AOX, classical plant thermogenin, and UCP, mammalian thermogenin,
to clarify the molecular mechanisms underlying thermoregulation in the skunk cabbage ( Symplocarpus renifolius ).
The SrAOX gene was first cloned and its sequence information was first revealed in this study.
Interestingly, this SrAOX transcript was specifically expressed in the thermogenic spadix, but not in the leaf, spathe and root.
The analyses with purified mitochondria from thermogenic florets showed that SrAOX protein was functionally expressed
in the mitochondria. Namely,
(1) SrAOX protein was detected using AOA monoclonal antibody in purified mitochondria,
(2) SrAOX can potentially exist as either a reduced or an oxidized dimer in a reversible manner
in vitro via the formation of disulphide bonds, (3) purified mitochondria, which express SrAOX,
were found to execute cyanide-insensitive respiration, and
(4) only pyruvate significantly stimulated SrAOX activity.
Furthermore, SrAOX protein existed as a non-covalently associated dimer in vivo.
Because it is known that two regulatory mechanisms of plant AOX, reduction state and α-keto acid interaction,
pyruvate concentration in the mitochondrial matrix or affinity to the SrAOX seems to be most important in regulating SrAOX activity. In summary, the experiments of thermal imaging in conjunction with expressional and biochemical analyses have shown that both AOX and UCP were functionally expressed in mitochondria isolated from thermogenic florets. Because neither of these protein expression levels significantly differed when ambient temperatures are changed, post-translational regulation of AOX and UCP would function to control respiration in response to such ambient temperature changes. The functional co-expression of AOX and UCP would significantly contribute to explosive, continuous and controlled respiration in the spadix. These results provide new insights into the mechanisms of thermoregulation in the skunk cabbage, Symplocarpus renifolius. The role of AOX and UCP, both of which are potentially thermogenic proteins in plant mitochondria, should be considered in future studies of thermogenic plants. (和訳) 植物における発熱は、内因性の熱産生によってある特定の花器官の温度が上昇する現象である。 この植物における発熱現象の観察は、フランスの生物学者である Jean-Baptiste de Lamarck による200年以上前の報告に端を発するが、 その後、サトイモ科のいくつかの種、ヤシ、ハス、ソテツそしてスイレンなど多くの植物における発熱現象が報告されている。 これまで調査されてきた発熱植物の中で、ハスとザゼンソウは恒温性を有する植物として知られている。 これらの植物は、花托や肉穂花序といった花器官の温度を外気温より上昇させることができ、さらに数日間その上昇させた体温を維持できる。 例えば、ハスは、外気温が10~45℃まで変化するような環境下においても、花托の温度を30~36℃に2~4日間維持することができる。 また、サトイモ科の植物であるザゼンソウは、外気温が0℃以下まで低下しても、 その肉穂花序の温度を熱産生により20℃内外に維持する能力を有する。 これまでの研究では、肉穂花序や花托といった発熱部位を対象とした様々な温度測定や呼吸測定が行われてきた。
しかしながら、植物における熱産生メカニズムの分子メカニズムは不明のままであった。
そこで本研究では、ザゼンソウ(Symplocarpus renifolius )における温度制御の基礎を成す分子メカニズムを
明らかにすることを目的として、従来から植物の発熱因子であると考えられてきたAOX(シアン耐性呼吸酵素;alternative oixdase)と
UCP(脱共役タンパク質;uncoupling protein)に着目した研究を展開した。 SrAOX 遺伝子は本研究により初めてクローン化され、シーケンス情報が公開された。
興味深いことに、SrAOX は発熱中の肉穂花序でのみ特異的に発現しており、葉、仏炎苞および根では発現が観察されなかった。
また、発熱中の肉穂花序・小花より精製したミトコンドリアを用いた解析から、SrAOXタンパク質はミトコンドリアにおいて
機能的に発現していることが明らかとなった。
すなわち、(1)精製したミトコンドリア画分では、AOAモノクローナル抗体に反応するSrAOXタンパク質の発現が観察された、
(2)in vitro においてSrAOXはジスルフィド結合を介して可逆的に還元型もしくは酸化型の二量体として存在し得る、
(3)、SrAOXが発現しているミトコンドリアの呼吸はシアン耐性呼吸を示した、
(4)ピルビン酸のみが有意にSrAOXを活性化した。さらに、in vivo ではSrAOXは活性化型である
非共有結合型(還元型)二量体として存在していた。
これらの結果は、SrAOXの活性調節には、ミトコンドリアのマトリックス内のピルビン酸濃度もしくはピルビン酸のSrAOXに対する
アフィニティーが最も重要な要因であると考えられる。 本研究では、赤外線画像解析とともに発現解析や生化学的解析を行い、 発熱組織である小花から精製したミトコンドリアにおいてAOXとUCPがともに機能的に発現していることを明らかにした。 また、外気温が変動した際でもこれらAOXやUCPのタンパク質の発現レベルが変化しないことから、 これらタンパク質の翻訳後修飾による活性制御が外気温変化に応答する呼吸制御に機能していると考えられた。 AOXとUCPの機能的共発現は、肉穂花序における爆発的かつ持続的な呼吸制御に貢献していると予想される。 本研究結果は、ザゼンソウにおける温度制御メカニズムに新たな知見をもたらすものであると考えられる。 今後は、植物においても発熱因子として機能し得るAOXとUCPの両方に着目した研究が成されることが期待される。 |
