学位論文題目 STUDIES ON MANIPULATION OF RUMINAL FERMENTATION AND METHANOGENESIS BY NATURAL PRODUCTS
( 天然物を用いるルーメン発酵及びメタン生成のマニピュレーションに関する研究 )
論文の内容の要旨

 The aims of the present studies were to investigate possible ways of manipulating ruminal fermentation in order to abate ruminal methanogenesis and reduce N excretion in feces and urine in ruminants by using safe natural products. The objectives of the studies were to investigate: (Expt.1) effects of Y. schidigera and Q. saponaria extracts and their combinations on in vitro ruminal fermentation and methane emissions; (Expt.2) effects of Y. schidigera and Q. saponaria , with or without galacto-oligosaccharides, on ruminal fermentation, CH4 production and N utilization in sheep; and (Expt.3) effects of Y. schidigera and Q. saponaria extracts on in vitro ruminal fermentation and methanogensis in adapted and unadapted rumen fluid.
Expt.1: YSE and QSE and their combinations may have the potential as beneficial ruminal fermentation manipulator with particular effects as antiprotozoal agents.
Expt.2: QSE and YSE reduces ruminal NH3-N and total VFA concentrations, and numerically decreases CH4 emissions, without having adverse effects on fiber digestion. In addition, administration of YSE, with or without GOS, numerically reduces protozoa numbers.
Expt.3: Supplementation with powdered Y. schidigera at 500 mg/day had no substantial effects on ruminal fermentation characteristics and protozoa numbers in sheep. Saponin-containing extracts of Q. saponaria and Y. schidigera may still favorably modify ruminal fermentation even when offered to adapted animals.

 本研究は、反芻動物からの温室効果ガス放出を低減することを目的とし、安全な天然物質を用いてルーメンメタン生成及び、 糞中及び尿中への窒素排出を低減させるためのマニピュレーションを明らかにする目的で次の3実験を実施した。 (実験1)in vitro ルーメン発酵及びメタン生成におよぼすYucca schidigera (YSE)、 Quillaja saponaria (QSE)の抽出液 及びそれらの併用添加の影響、 (実験2)めん羊のルーメン発酵、メタン生成及び窒素代謝におよぼすYSE及び QSEとガラクトオリゴ糖の併用添加の効果  (実験3)ユッカ粗製粉末(PYE)給与に馴致しためん羊、或いは馴致していないめん羊のルーメン液における in vitroルーメン発酵及びメタン生成におよぼすYSE及びQSEの影響。
 以上の実験結果からYucca schidigera 及びQuillaja saponaria 抽出液がプロトゾアに対して強力な抑制作用を有し、 ルーメンアンモニア及びメタン生成の低減に関連したと考えられる。 また、Yucca schidigera 及びQuillaja saponaria 抽出液の併用は単独の場合よりも再サポニン化合物の特色が発揮され、 より有効なルーメンマニピュレータになり得る可能性がある。 さらにガラクトオリゴ糖との併用によって両サポニン化合物は安全で有効なルーメン発酵マニピュレータになり得る。 これらのサポニンの効果は馴致によって強化される可能性が示唆された。
